COMPARISON OF 2,2’-DITHIODIPYRIDINE AND 5,5’-DITHIOBIS-2-NITROBENZOIC ACID IN DOG WHOLE BLOOD CHOLINESTERASE DETERMINATIONS
Abstract
The use of 2,2’-dithiodipyridine (2-PDS) as chromophore for cholinesterase determination in dog whole blood is evaluated and compared with ELLMAN chromophore 5,5’-dithiobis-2-nitrobenzoic acid (DTNB). Good precision was obtained using both chromophores (within and between-run CVs < 8%). However, higher cholinesterase values were obtained when 2-PDS was used. The use of more concentrated whole blood dilution (1:20 dilution ratio) was only allowed using 2-PDS as chromophore. Ability of 2-PDS to avoid haemoglobininterference at 340 nm wavelenght could explain these results. Both chromophores gave similar background ratio. Similar inhibition was detected using both 2-PDS and DTNB in 1:50 whole blood dilution ratio after “in vivo” and “in vitro” organophosphate coumaphos exposure (correlation coefficients between chromophores > 0.90). The 1:20 whole blood dilution, assayed with 2-PDS, yielded similar inhibition results than 1:50. Therefore, these results recommend the use of 2-PDS as chromophore for cholinesterase determination in dog whole blood samples.Downloads
