Abstract

Cells of Escherichia coli, strains K12 (rec+) and HBlOl (recA-), carrying the recombinant plasmid pTG201 were immobilized in order to study the stability and expression of plasmid genes in several culture conditions. Continuous cultures (chemostats) with BL medium and different oxygen tension were tested. Parallel experiments were carried out with free cells. The statistical analysis of the kinetics of plasmid loss showed a better plasmid stability in immobilized cells cultured in the presence of pure oxygen. In the last case a maximal production of protein codified by plasmid also occur. Likewise the influence of culture conditions over the microbial growth in the gel and their relation to plasmid stability has also been studied.
