Clorofilasa en hojas de citrus. I. Extracción, medida y factores que influyen en la actividad del enzima

Resumen

Different ways of preparation of the material containing the enzyme and for determination of chlorophyllase activity in homogeneus and heterogeneus media, in Citrus Limón (Vema variety) are studied. The way to prepare the enzymatic material is that of «acetonic powder» and in order to determine its activity an homogeneus médium has been used, in which the enzyme has been extracted with the buffered media: Tris 0,041 M — Glycin 0,32 M—Glucose 0,70 M + Dodecyl hydrogen sulfate (sodium salt) 0,5 %, pH = 8,50. The activity ís expressed as % of hydrolised chlorophylles in the reaction time. At the same time, several factors of the reaction media influencing on the activity, such as: acetone concentration, citrate concentration, pH and temperatura, are studied.
